NK细胞具有非特异性杀伤肿瘤细胞的活性,并有监视肿瘤发生和防止肿瘤转移的作用。中药是癌症治疗方法中的一个重要组成部分。本文以绞股蓝、黄芪为主的中药方剂对泌尿生殖系肿瘤患者进行治疗并观察其NK细胞活性改变。 1 材料与方法 1.1 NK细胞活性检测:参照王甫厚的乳酸脱氢酶释放法检测外周血NK细胞活性。取抗凝血3ml分离出单个核细胞做为效应细:胞,靶细胞系传代培养24小时的K562细胞,效应细胞与靶细胞比例10∶1,置37℃5％二氧化碳孵箱培养24小时,在酶联免疫检测仪490nm处读取光密度值,计算NK细胞杀伤活性以杀伤K562细胞百分率表示。
NK cells have the activity of non-specific killing of tumor cells and have the effect of monitoring tumorigenesis and preventing tumor metastasis. Traditional Chinese medicine is an important part of cancer treatment. In this article, the traditional Chinese medicine prescriptions of Gynostemma pentaphyllum and Astragalus membranaceus were used to treat patients with genitourinary neoplasms and observe their NK cell activity changes. 1 Materials and methods 1.1 NK cell activity assay: The NK cell activity in peripheral blood was measured with reference to Wang Qiang’s lactate dehydrogenase release assay. Take anticoagulant 3ml to isolate mononuclear cells as effector cells: cells and target cell lines were subcultured for 24 hours in K562 cells, the ratio of effector cells to target cells was 10:1, and incubated at 37°C in a 5% carbon dioxide incubator for 24 hours. Optical density values were read at 490 nm in an enzyme-linked immunosorbent assay and the killing activity of NK cells was calculated to represent the percentage of K562 cells.
