目的观察2型糖尿病(T2DM)大鼠逼尿肌细胞线粒体超微结构的改变。方法建立T2DM大鼠动物模型,利用透射电镜观察其逼尿肌细胞内线粒体结构并应用BI2000序列图像分析软件检测线粒体密度。结果对照组的线粒体相对光密度为2.417±0.376,相对灰度为0.572±0.083;T2DM组逼尿肌细胞内线粒体相对光密度为1.549±0.871,相对灰度为0.836±0.279,线粒体基质密度较正常明显下降。结论糖尿病大鼠逼尿肌细胞内线粒体超微结构破坏,基质密度下降,提示逼尿肌细胞能量代谢障碍是糖尿病膀胱收缩力降低的原因。
Objective To observe the changes of mitochondrial ultrastructure in detrusor muscle cells of type 2 diabetes mellitus (T2DM) rats. Methods The animal model of T2DM was established. The mitochondrial structure of detrusor cells was observed by transmission electron microscopy. The density of mitochondria was detected by BI2000 sequence analysis software. Results The relative optical density of mitochondria in control group was 2.417 ± 0.376 and the relative gray level was 0.572 ± 0.083. The relative optical density of mitochondria in detrusor muscle cells of T2DM group was 1.549 ± 0.871, the relative gray level was 0.836 ± 0.279, and the mitochondrial matrix density was normal Significant decline. Conclusion The ultrastructure of mitochondria in detrusor cells of diabetic rats is destroyed and the matrix density is decreased, suggesting that the energy metabolism disorder of detrusor cells is the reason of the decrease of bladder contractility in diabetic rats.
