[目的]探讨丹皮酚(paeonol,Pae)和顺铂(CDDP)联合用药对人宫颈癌SiHa细胞的增殖抑制、凋亡诱导作用。[方法]应用MTT法检测不同浓度的Pae或CDDP单独及联合用药对宫颈癌SiHa细胞增殖的抑制作用,同时观察Pae与CDDP的协同抗肿瘤作用。流式细胞仪测定Pae联合CDDP用药对SiHa细胞凋亡的诱导作用。[结果]各浓度Pae(9.375~300mg/L)和CDDP(0.625~10mg/L)对人宫颈癌SiHa细胞均有明显增殖抑制作用,且呈时间—剂量依赖效应关系。18.75、37.5、75mg/LPae分别与2.5、5、75mg/LCDDP联用时具有协同作用,且以75mg/LPae与5mg/LCDDP联用时协同作用最显著(CDI=0.544)。75mg/LPae组细胞凋亡率为8.26%±1.12%,5mg/LCDDP组细胞凋亡率为29.62%±2.48%,与75mg/LPae联合5mg/LCDDP组细胞凋亡率(52.49%±4.39%)比较均有显著性差异(P<0.01)。[结论]Pae与CDDP联合应用具有显著的协同抗人宫颈癌SiHa细胞增殖作用,其机制可能与两者协同诱导SiHa细胞凋亡有关。
[Objective] To investigate the inhibitory effect of paeonol (Pae) and cisplatin (CDDP) on the proliferation and apoptosis of human cervical cancer SiHa cells. [Method] The inhibitory effect of different concentrations of Pae or CDDP alone or in combination on the proliferation of cervical cancer SiHa cells was detected by MTT assay. The synergistic anti-tumor effect of Pae and CDDP was also observed. Induction of apoptosis in SiHa cells by Pae combined with CDDP by flow cytometry. [Result] Pae (9.375 ~ 300 mg / L) and CDDP (0.625 ~ 10 mg / L) had significant inhibitory effects on SiHa cells in a time-and dose-dependent manner. The synergistic effects were observed when combined with 2.5, 5 and 75mg / LDPDP at 18.75, 37.5 and 75mg / LPae, respectively. The synergistic effect was most significant when combined with 75mg / LPae and 5mg / LCDDP (CDI = 0.544). The rate of apoptosis in the group of 75mg / LPae was 8.26% ± 1.12%, the rate of apoptosis in the group of 5mg / LCDDP was 29.62% ± 2.48%, the rate of apoptosis was 52.49% ± 4.39% in combination with 75mg / LPae 5mg / There were significant differences (P <0.01). [Conclusion] The combination of Pae and CDDP has a significant synergistic effect on the proliferation of SiHa cells, which may be related to the synergistic effect of Pae and CDDP on SiHa cell apoptosis.
