目的:为准确测定虫草类相关保健品中腺苷的含量,对高效液相色谱法测定腺苷进行了优化。方法:采用10%甲醇水溶液作为提取液,密塞超声30 min,以Supelco C18(250 mm×4.6 mm,5μm)为色谱分析柱,磷酸盐缓冲液(pH6.5)+甲醇(90+10)为流动相洗脱,流速为1.0 ml/min,检测波长为260 nm进行测定。结果:腺苷在12.8 ng~128 ng范围内呈良好的线性,相关系数1.000,方法的回收率为99.7%~99.9%,方法精密度RSD为0.93%。结论:该方法灵敏度高、选择性好,线性关系及重现性好,适合于虫草类相关保健品中活性成分腺苷的含量分析。
Objective: To determine the content of adenosine in Cordyceps related health products accurately, the adenosine was determined by HPLC. Methods: A 10% methanol aqueous solution was used as the extract. The mixture was sonicated for 30 min with a Supelco C18 (250 mm × 4.6 mm, 5 μm) column. Phosphate buffer (pH 6.5) + methanol The mobile phase was eluted at a flow rate of 1.0 ml / min with a detection wavelength of 260 nm. Results: Adenosine showed a good linearity in the range of 12.8 ng ~ 128 ng with a correlation coefficient of 1.000. The recovery rate of the method was 99.7% -99.9%. The precision of the method was 0.93%. Conclusion: This method has high sensitivity, good selectivity, good linearity and good reproducibility. It is suitable for the content analysis of adenosine in active ingredients of Cordyceps related health products.
