目的:研究猫爪草醇提取物的抗耐多药结核分枝杆菌(multiple drugs resistant bacillus tuberculosis,MDR-TB)作用。方法:采用MDR-TB菌液灌胃制备小鼠MDR-TB感染病理模型,将30只健康雄性小鼠随机分为3组,正常组、模型组和猫爪草提取物组,每组10只,采用ELISA法检测小鼠血清中与结核细胞免疫密切相关的细胞因子IFN-γ、IL-4、IL-10和IL-12含量的变化,同时分离出单个核细胞,抽提全部RNA,采用RT-PCR法检测单个核细胞中IFN-γ、IL-4、IL-10、IL-12及GLS的mRNA变化。结果:猫爪草提取物组小鼠血清中的IFN-γ为(2.0±0.7)pg.mL-1、IL-4为(6.0±1.5)pg.mL-1、IL-10为(12.1±3.1)pg.mL-1、IL-12为(3.0±0.9)pg.mL-1,与模型组比较,IFN-γ、IL-12含量明显增强,IL-4、IL-10含量明显降低(P<0.05或P<0.01)。在mRNA表达水平,IFN-γ、IL-12、GLS表达明显增强,IL-4、IL-10 mRNA表达明显降低(P<0.05或P<0.01)。结论:猫爪草醇提取物可以通过上调基因转录水平增强小鼠的细胞免疫功能,具有较显著的抗MDR-TB作用。
Objective: To study the anti-multi-drug resistant bacillus tuberculosis (MDR-TB) effect of the extract of Radix Ranunculus. Methods: The pathological model of MDR-TB infection in mice was prepared by gavage with MDR-TB. Thirty healthy male mice were randomly divided into three groups: normal group, model group and cat’s paw extract group, with 10 rabbits in each group , The contents of cytokines IFN-γ, IL-4, IL-10 and IL-12 in the serum of the mice that were closely related to the immunity of the TB were detected by ELISA. Meanwhile, the mononuclear cells were isolated and all the RNA was extracted. The mRNA expressions of IFN-γ, IL-4, IL-10, IL-12 and GLS in mononuclear cells were detected by RT-PCR. Results: The serum levels of IFN-γ in the extract of cat litter group were (2.0 ± 0.7) pg · mL-1, (6.0 ± 1.5) pg · mL-1 for IL-4 and 12.1 ± 3.1) pg.mL-1 and IL-12 were (3.0 ± 0.9) pg.mL-1 respectively. Compared with the model group, the contents of IFN-γ and IL-12 were significantly increased and the levels of IL-4 and IL- P <0.05 or P <0.01). At mRNA level, the expression of IFN-γ, IL-12 and GLS were significantly increased, while the expressions of IL-4 and IL-10 mRNA were significantly decreased (P <0.05 or P <0.01). Conclusion: The extract of Radix Ranunculus could enhance the cellular immune function of mice by up-regulating gene transcription level, and has a significant anti-MDR-TB effect.
