目的:探讨组蛋白去乙酰基酶抑制剂丁酸钠对卵巢癌细胞株CAOV3侵袭能力的抑制作用及其对血管内皮生长因子(VEGF)表达的影响。方法:用不同浓度的丁酸钠处理CAOV3细胞,采用锥虫蓝活细胞拒染法检测癌细胞体外生长活性,Transwell小室观察其对细胞运动及侵袭能力的影响,利用RT-PCR技术检测CAOV3细胞中VEGF mRNA表达的变化。结果:丁酸钠抑制CAOV3细胞的生长,并呈剂量依赖性(P<0.05);其对CAOV3细胞的运动及侵袭能力均有明显抑制作用(P<0.05);并且能显著降低VEGF mRNA的表达(P<0.05)。结论:丁酸钠能有效抑制卵巢癌细胞株CAOV3的侵袭转移,可能与降低VEGF的表达有关。
AIM: To investigate the inhibitory effect of sodium butyrate, a histone deacetylase inhibitor, on the invasiveness of ovarian cancer cell line CAOV3 and its effect on the expression of vascular endothelial growth factor (VEGF). Methods: The CAOV3 cells were treated with different concentrations of sodium butyrate. The growth activity of cancer cells was detected by trypan blue exclusion method. The transwell chamber was used to observe the effect on cell motility and invasion. The expression of CAOV3 cells In the changes of VEGF mRNA expression. Results: Sodium butyrate inhibited the growth of CAOV3 cells in a dose-dependent manner (P <0.05), and inhibited the migration and invasion of CAOV3 cells (P <0.05), and significantly reduced the expression of VEGF mRNA (P <0.05). CONCLUSION: Sodium butyrate can effectively inhibit the invasion and metastasis of ovarian cancer cell line CAOV3, which may be related to the decrease of VEGF expression.
