目的 了解甘肃裕固族人群 D3 S13 5 8、D16S5 3 9、TH0 1、TPOX、CSF1PO、D7S82 0六个 STR位点遗传多态性。 方法 随机抽取 10 3名甘肃裕固族无血缘关系个体的静脉血 ,应用 Am p Fl STR Cofiler试剂盒扩增 6个 STR位点 ,PCR产物经 ABI Prism 3 77序列分析仪电泳后 ,用基因扫描软件进行分析。 结果 6个 STR位点均符合 Hardy-Weinberg平衡 ,D3 S13 5 8、D16S5 3 9、TH0 1、TPOX、CSF1PO、D7S82 0位点的杂合度分别为 0 .82 99,0 .93 84,0 .885 1,0 .7746,0 .85 17,0 .943 6,期望排除率分别为 0 .44 5 6,0 .60 2 8,0 .5 0 78,0 .42 67,0 .5 3 40 ,0 .60 47,个体识别率分别为 0 .6916,0 .782 0 ,0 .70 81,0 .64 5 5 ,0 .745 2 ,0 .7863。累积 DP为 0 .9996,累积 PE为 0 .9886,累积 PIC为 0 .9994。 结论 裕固族群体有其自身的 STR等位基因分布特征。所获数据可为法医学个体识别、亲子鉴定及遗传学研究提供依据。
Objective To understand the genetic polymorphisms of six STR loci in D3 S13 5 8, D16S5 3 9, TH0 1, TPOX, CSF1PO and D7S82 0 in Yugur people in Gansu Province. Methods A total of 10 venous blood samples were collected from Yumou unrelated individuals in Gansu Province. Six STR loci were amplified by Am p Fl STR Cofiler kit. The PCR products were electrophoresed by ABI Prism 3 77 sequencer and sequenced by Gene Scanning Software Analyze. Results All six STR loci were in accordance with Hardy-Weinberg equilibrium. The heterozygosities of D3 S13 5 8, D16S5 3 9, TH0 1, TPOX, CSF1PO and D7S82 0 were 0.82 99,0 .93 84,0 respectively. 885 1,0 .7746,0 .85 17,0 .943 6, the expected exclusion rates were 0 .44 5 6,0 .60 2 8,0 .5 0 78,0 .42 67,0 .5 3 40 , 0 .60 47, and the individual recognition rates were 0.6916, 0.782 0, 0.70 81, 0.64 5 5, 0. 745 2, 0.7863 respectively. The cumulative DP is 0.9996, the cumulative PE is 0.9886, and the accumulated PIC is 0.9994. Conclusion The Yugur people have their own distribution of STR alleles. The data obtained for forensic individual identification, paternity testing and genetic studies provide the basis.
