目的观察新疆阿魏挥发油(FSEO)对小鼠的镇痛作用以及对吗啡依赖大鼠、小鼠戒断症状的影响。方法采用扭体法、热板法观察FSEO对小鼠的镇痛作用,吗啡依赖大鼠、小鼠采用纳洛酮催促戒断,观察FSEO对戒断症状的影响。结果灌服不同剂量FSEO可不同程度的抑制化学刺激所致的小鼠扭体反应,提高热板致痛小鼠的痛阈值,扭体镇痛的ED50=3.26 g(生药量).kg-1。FSEO各剂量组对吗啡依赖大鼠催促戒断症状均有一定的抑制作用,还可明显抑制催促戒断所致的大鼠体重减轻,其体重下降与模型对照组比较有统计学。FSEO各剂量组对纳洛酮催促戒断后吗啡依赖小鼠的动物跳跃反应有一定程度的减少,而对催促戒断后小鼠的体重下降有较好的抑制作用,与模型对照组比较均有统计学差异。结论 FSEO对小鼠有一定的镇痛作用,对纳洛酮催促戒断后吗啡依赖大鼠、小鼠的体重下降和戒断症状有较好的抑制作用。提示FSEO作为戒毒新药有良好的研究价值和应用前景。
Objective To observe the analgesic effect of volatile oil of Ferulicus vulgaris (FSEO) on mice and its effect on the withdrawal symptoms of morphine-dependent rats and mice. Methods The writhing method and hot plate method were used to observe the analgesic effect of FSEO on mice. Morphine-dependent rats and mice were induced to withdraw with naloxone to observe the effect of FSEO on withdrawal symptoms. Results Different doses of FSEO could inhibit the writhing reaction of mice induced by chemical stimulation and increase the pain threshold of hot-plate pain-induced mice. The ED50 of writhing analgesia was 3.26 g (crude drug amount) .kg-1 . Each dose group of FSEO had some inhibitory effects on withdrawal symptoms of morphine-dependent rats, but also significantly inhibited the weight loss of rats induced by withdrawal, and its weight loss was statistically compared with the model control group. FSEO dose groups of morphine-dependent mice after withdrawal of naloxone to urge the animal’s jump reaction to a certain extent reduce, but to urge the withdrawal of mice after weight loss had a better inhibitory effect, compared with the model control group There are statistical differences. Conclusion FSEO has certain analgesic effect on mice and good inhibition of weight loss and withdrawal symptoms of morphine-dependent rats and mice after withdrawal of naloxone. It is suggested that FSEO has good research value and application prospect as a new drug for detoxification.
