我们提出一种新型非标记蛋白质的电化学检测转录因子NF-κB的方法,即将固定在金电极上的通用寡核苷酸通过连接酶形成单分子发夹寡核苷酸,然后进行固相酶促延伸成双链寡核苷酸,最后通过Eco R的酶切信号的变化,采用阳极溶出法和循环伏安法结合纳米金催化银沉积法检测NF-κB。结果表明本方法具有高特异性、高灵敏度和快速等优点,为单核苷酸突变、药物筛选研究提供有力工具。
