目的:研究人端粒酶逆转录酶(human telom erase reverse transcriptase,hTERT)与P糖蛋白与bcl2蛋白在胰腺癌组织中的表达及其关系和临床意义。方法:用链霉抗生物素蛋白生物素过氧化物酶复合物法检测hTERT,链霉抗生物素蛋白过氧化物酶法检测P糖蛋白,链霉抗生物素蛋白生物素标记法检测bcl2蛋白,并用Pearson关联系数(rp)研究彼此的关联性。结果:在胰腺癌组织中hTERT的阳性率为82%,而在正常胰腺组织和慢性胰腺炎组织中无表达(均为P<0.001);P糖蛋白在胰腺癌组织中表达的阳性率为53%,明显高于慢性胰腺炎组织、正常胰腺组织的22%、17%(均为P<0.05);bcl2蛋白在胰腺癌组织中表达的阳性率为58%,而正常胰腺组织和慢性胰腺炎组织均无表达(均为P<0.001)。在胰腺癌组织中,hTERT的表达分别与P糖蛋白和bcl2蛋白的表达有关联性(P<0.05,P<0.001);P糖蛋白和bcl2蛋白的表达也有关联性(P<0.001)。结论:hTERT、P糖蛋白与bcl2蛋白在胰腺癌组织中均有较高的表达,hTERT与P糖蛋白和bcl2蛋白的表达分别具有关联性,提示hTERT在胰腺癌细胞中具有耐药和抗凋亡的作用,检测hTERT和bcl2蛋白有助于鉴别胰腺癌和慢性胰腺炎,而且可通过检测hTERT来预测化学治疗的效果。
OBJECTIVE: To study the expression of human telomerase reverse transcriptase (hTERT), P-glycoprotein and bcl2 protein in pancreatic cancer and their clinical significance. Methods: hTERT was detected by streptavidin biotin peroxidase complex method, P glycoprotein was detected by streptavidin peroxidase method, bcl2 protein was detected by streptavidin biotin labeling method And Pearson correlation coefficient (rp) to study each other’s relevance. Results: The positive rate of hTERT in pancreatic cancer was 82%, but not in normal pancreatic tissue and chronic pancreatitis (all P <0.001). The positive rate of P-glycoprotein expression in pancreatic carcinoma was 53 %, Which was significantly higher than those in chronic pancreatitis and normal pancreatic tissues (22% and 17% respectively) (all P <0.05). The positive rate of bcl2 protein expression in pancreatic cancer was 58%, while that in normal pancreatic tissue and chronic pancreatitis Tissues were not expressed (all P <0.001). In pancreatic cancer tissues, the expression of hTERT was correlated with the expression of P-glycoprotein and bcl2 protein, respectively (P <0.05, P <0.001). The expressions of P-glycoprotein and bcl2 were also correlated with each other (P <0.001). CONCLUSIONS: The expression of hTERT, P-glycoprotein and bcl2 protein in pancreatic cancer tissues are high, hTERT is associated with the expression of P-glycoprotein and bcl2 protein respectively, suggesting that hTERT is resistant and resistant to apoptosis in pancreatic cancer cells The role of death, detection of hTERT and bcl2 protein helps identify pancreatic cancer and chronic pancreatitis, but also by measuring hTERT to predict the effect of chemotherapy.
