目的:探讨survivin和PTEN在非小细胞肺癌(NSCLC)中的表达情况及其与NSCLC临床生物学行为的关系。方法:应用免疫组织化学方法检测survivin和PTEN在78例NSCLC组织、46例癌旁组织和12例非癌肺良性病变组织中的表达情况,并将结果进行相关性分析。结果:Survivin在非癌良性病变组织、癌旁组织、非小细胞肺癌中的阳性表达率分别是0、10.9%、67.9%,其蛋白表达逐渐增强(P<0.05);非小细胞肺癌中的survivin表达显著高于癌旁肺组织及非癌良性病变组织中的表达(P<0.05)。PTEN在非癌良性病变组织、癌旁组织、非小细胞肺癌中的阳性表达率分别是100%、91.3%、47.4%,其表达逐渐减弱(P<0.05);非小细胞肺癌中的PTEN表达低于癌旁肺组织及非癌良性病变组织中的表达(P<0.05)。survivin蛋白的表达与NSCLC的病理学分级、淋巴结转移、TNM分期有关(P<0.05)。PTEN蛋白的表达与NSCLC的病理学分级、淋巴结转移、TNM分期有关(P<0.05)。Survivin与PTEN表达呈负相关(P<0.05)。结论:Survivin和PTEN在NSCLC中的异常表达与其生物学行为密切相关,survivin在非小细胞肺癌中过度表达,PTEN在非小细胞肺癌中表达降低,并且二者的表达呈负相关,二者可作为NSCLC判断病情和评价预后的参考指标。
Objective: To investigate the expression of survivin and PTEN in non-small cell lung cancer (NSCLC) and its relationship with clinical biological behavior of NSCLC. Methods: The expressions of survivin and PTEN were detected by immunohistochemistry in 78 cases of NSCLC, 46 cases of paracancerous tissues and 12 cases of benign non-cancerous lung tissues. The correlations were analyzed. Results: The positive rates of Survivin in non-cancerous lesions, paracancerous tissues and non-small cell lung cancer were 0, 10.9% and 67.9%, respectively, and their protein expressions were gradually increased (P <0.05). In non-small cell lung cancer Survivin expression was significantly higher than that in paracancer lung tissue and non-cancerous benign lesions (P <0.05). The positive rates of PTEN expression in non-cancerous benign lesions, paracancerous tissues and non-small cell lung cancer tissues were 100%, 91.3% and 47.4%, respectively. The expression of PTEN was decreased (P <0.05). PTEN expression in non-small cell lung cancer Which was lower than that in paracancer tissues and benign non-cancerous tissues (P <0.05). The expression of survivin protein was correlated with pathological grade, lymph node metastasis and TNM staging of NSCLC (P <0.05). The expression of PTEN protein was correlated with pathological grade, lymph node metastasis and TNM staging of NSCLC (P <0.05). Survivin and PTEN expression was negatively correlated (P <0.05). CONCLUSIONS: The abnormal expression of Survivin and PTEN in NSCLC is closely related to their biological behavior. Survivin is over-expressed in non-small cell lung cancer and PTEN is decreased in non-small cell lung cancer, and the expression of both is negatively correlated As a reference to judge the condition of NSCLC and evaluate the prognosis.
