本文运用Westerm blot及抗体介导的特异性细胞增殖试验观察抗HER2/neu胞外区不同表位的单克隆抗体对SKBR3乳腺癌细胞株酪氨酸磷酸化及细胞增殖的影响.实验结果发现单抗c-neu-5对SKBR3细胞P185蛋白酪氨酸磷酸化有明显促进作用,而对SKBR3细胞增殖作用不明显;单抗c-neu-2对SKBR3细胞酪氨酸磷酸化及细胞增殖均有明显抑制作用,对细胞增殖的抑制率可达49.8%.
In this study, Westerm blot and antibody-mediated cell proliferation assays were used to observe the effect of monoclonal antibodies against different epitopes of HER2/neu extracellular domain on tyrosine phosphorylation and cell proliferation in SKBR3 breast cancer cell lines. Anti-c-neu-5 significantly promoted the tyrosine phosphorylation of P185 protein in SKBR3 cells, but had no obvious effect on the proliferation of SKBR3 cells. Monoclonal antibody c-neu-2 had tyrosine phosphorylation and cell proliferation in SKBR3 cells. Significant inhibition, inhibition of cell proliferation up to 49.8%.
