目的白细胞介素17在多种心血管疾病中如自身免疫性心肌炎、病毒性心肌炎、扩张型心肌病、不稳定型冠心病等表达增加,在心血管病慢性炎症性过程起到了一定作用。分析白细胞介素17在慢性心力衰竭大鼠心肌中的表达特点。方法采用腹主动脉缩窄术构建大鼠慢性心力衰竭模型。将62只雄性SD慢性心力衰竭大鼠模型随机分14d组、28d组、56d组及112d组,另设假手术组大鼠10只。采用酶联免疫吸附测定法(ELISA)、免疫荧光激光共聚焦技术(LSCM)及蛋白免疫印迹法(Western blot)检测各组大鼠心肌IL-17的表达。结果造模成功率为58.06%。ELISA、Western blot、激光共聚焦的结果均表明在大鼠慢性心力衰竭的发生发展过程中,IL-17在心肌组织大量表达,且随着时间变化而逐渐升高,在112d表达最为显著。结论 IL-17作为一种强力的促炎症因子在慢性心力衰竭大鼠心肌组织中大量表达,可能与慢性心力衰竭后的心室重塑及心律失常的发生有关。
Objective Interleukin-17 is expressed in many cardiovascular diseases such as autoimmune myocarditis, viral myocarditis, dilated cardiomyopathy, and unstable coronary heart disease. It plays a role in the chronic inflammatory process of cardiovascular disease. Analysis of the expression of interleukin 17 in the myocardium of chronic heart failure rats. Methods A rat model of chronic heart failure was established by abdominal aortic constriction. 62 male SD rats with chronic heart failure were randomly divided into 14d group, 28d group, 56d group and 112d group, another 10 rats were sham operated group. The myocardial IL-17 expression in each group was detected by enzyme-linked immunosorbent assay (ELISA), immunofluorescence confocal laser scanning microscopy (LSCM) and Western blot. Results The success rate of modeling was 58.06%. The results of ELISA, Western blot and confocal laser scanning showed that IL-17 was highly expressed in myocardium during the development of chronic heart failure in rats, and gradually increased with time. The expression of IL-17 was the most significant at 112 days. CONCLUSION: IL-17 is a potent proinflammatory cytokine expressed in myocardial tissue of rats with chronic heart failure, which may be related to ventricular remodeling and arrhythmia after chronic heart failure.
