为评价2015年湖北省水稻区试两系不育系材料的遗传相似性并构建其指纹图谱,利用48个SSR分子标记对15份参试品种及6个代表性对照品种进行分析,筛选出12对扩增稳定、多态性好并分布均匀的SSR引物,共检测到47个等位基因(Na),每个标记平均检测到3.9个,每标记多态性信息含量(PIC)平均值为0.64,利用这12个引物为核心标记构建了供试材料的指纹图谱;聚类分析结果表明,15份参试品种与6个对照品种在相似系数0.66左右分为6个类群,其中6个品种与广占63-4S均归于三类群,占参试品种的40%,此外,四类群的3个品种还与三类群同属一个亚类群;其余6个品种分别与Y58S、农垦58S以及珍汕97A归于同一类群。因此,聚类分析结果表明参试品种表现出了背景来源单一、亲缘关系较近的趋势。本研究结果有助于湖北省水稻两系不育系资源利用、创新与引进,持续提高水稻育种水平。
In order to assess the genetic similarity of CMS lines in Hubei province in 2015 and to construct their fingerprints, 48 SSR markers were used to analyze 15 tested cultivars and 6 representative control cultivars, and 12 A total of 47 alleles (Na) were detected for SSR primers with stable amplification, good polymorphism and uniform distribution. The average number of each marker was 3.9, and the average value of polymorphism information content (PIC) per marker was 0.64. The 12 primers were used as the core markers to construct the fingerprint of the tested materials. The cluster analysis showed that 15 tested cultivars and 6 control cultivars were divided into 6 groups at the similarity coefficient of 0.66, of which 6 cultivars And widely accounted for 63-4S are attributed to three groups, accounting for 40% of the varieties tested, in addition, the four groups of three varieties also belong to the same three sub-groups and a group of the other six varieties were Y58S, Nongken 58S and Zhenshan 97A Attributed to the same group. Therefore, the cluster analysis showed that the tested varieties showed a single source with a relatively close genetic relationship. The results of this study contribute to the utilization, innovation and introduction of two-line CMS lines in Hubei Province, and continuously improve the level of rice breeding.
