目的观察三七总皂苷(PNS)联合雷公藤多苷(TG)对胶原诱导关节炎(CIA)大鼠外周血TNF-α、IL-6、PAI-1水平以及心肌MCP-1表达的影响。方法将复制成功的CIA大鼠随机分为模型组、TG(10mg/Kg.d)组、PNS(200mg/Kg.d)+TG(10mg/Kg.d)组和MTX(1.6mg/Kg.w)组,另设正常大鼠为空白组,给药6周后采血,开胸取心脏。采用液相蛋白芯片技术检测血清TNF-α、IL-6、PAI-1含量,免疫组织化学法检测心肌MCP-1蛋白表达。结果与正常对照组比较,CIA模型组大鼠血清TNF-α、IL-6、PAI-1水平明显升高,心肌MCP-1蛋白表达显著上调;TG联合PNS可明显降低血清中TNF-α、IL-6、PAI-1水平,改善心肌的病理状态,下调MCP-1在心肌中的表达,其作用显著优于TG和MTX组。结论中药有效成分TG联合PNS对心血管损伤有显著防治作用,其机制可能与降低血清TNF-α、IL-6、PAI-1及心肌MCP-1表达从而发挥抗炎作用有关。
Objective To observe the effect of Panax Notoginseng Saponins (PNS) combined with Tripterygium glycosides (TG) on the levels of TNF-α, IL-6 and PAI-1 in peripheral blood and the expression of MCP-1 in myocardium induced by collagen-induced arthritis in rats. Methods CIA rats with successful replication were randomly divided into model group, TG (10mg / Kg.d) group, PNS (200mg / Kg.d) + TG (10mg / Kg.d) group and MTX (1.6mg / Kg. w) group, another normal rats as a blank group, 6 weeks after administration of blood, chest to take the heart. The contents of TNF-α, IL-6 and PAI-1 in serum were detected by liquid protein chip technique. The expression of MCP-1 in myocardium was detected by immunohistochemistry. Results Compared with the normal control group, the levels of serum TNF-α, IL-6 and PAI-1 in the CIA model group were significantly increased and the expression of MCP-1 protein in the myocardium was significantly increased. TG combined with PNS significantly decreased the levels of TNF- IL-6, PAI-1 levels, improve the pathological state of myocardium and downregulate the expression of MCP-1 in myocardium, its effect is significantly better than TG and MTX group. Conclusion TG combined with PNS has a significant preventive and therapeutic effect on cardiovascular injury. The mechanism may be related to the decrease of serum TNF-α, IL-6, PAI-1 and MCP-1 in myocardium.
