目的 观察维生素A(VA)对碘过量小鼠肝脏胰岛素样生长因子-Ⅰ(IGF-Ⅰ)表达的影响.方法 昆明种小鼠按体质量随机分成6组:对照组(含碘50 μg/L,VA 4000 U/kg);高碘组(含碘3000 μg/L,VA 4000 U/kg);常碘低VA组(含碘50 μg/L,VA 0 U/kg);高碘低VA组(含碘3000 μg/L,VA 0 U/kg);高碘补VA1组(含碘3000 μg/L,VA 8000 U/kg);高碘补VA2组(含碘3000 μg/L,VA 16 000 U/kg).喂养3、6个月时,ELISA法检测小鼠血清IGF-Ⅰ水平,RT-PCR法检测肝脏IGF-Ⅰ mRNA表达水平.结果 与对照组相比,3个月时各组血清IGF-Ⅰ水平未见明显改变,6个月时高碘组、常碘低VA组、高碘低VA组血清IGF-Ⅰ水平明显下降(q=3.52、3.29、3.84,P<0.05),高碘补VA组,IGF-Ⅰ水平有所升高,介于对照组和高碘组之间.3、6个月时,高碘和常碘低VA组肝脏IGF-Ⅰ mRNA表达水平,与对照组相比,均有降低趋势;3个月时高碘补VA组与高碘组比较,肝脏IGF-Ⅰ mRNA表达有升高趋势,而6个月时,则未见明显改变,经检验各组差异均无统计学意义(P>0.05).结论 高碘摄入抑制小鼠肝脏合成分泌IGF-Ⅰ,补充VA可以部分拮抗高碘的抑制作用.
