本文报导应用气质联用(GC—MS)、高效液相色谱(HPLC)、薄层扫描(TLCS)等法研究青蒿酯钠的命运。实验结果提示:青蒿酯钠在体内外可转化为双氢青蒿素,双氢青蒿素可视为青蒿酯钠的有效代谢物,其在犬体内动力学为一级速率反应,具有一房室模型特征,半衰期i_(1/2)10分钟,表现分布容积Vd 0.36LL/kg。青蒿酯钠等药物具有诱导肝药酶的作用,首过效应明显。
This paper reports the use of gas chromatography-mass spectrometry (GC-MS), high performance liquid chromatography (HPLC), thin layer scanning (TLCS), etc. to study the fate of sodium artemisinate. The experimental results suggest that: Artemisinin sodium can be converted into dihydroartemisinin in vitro and in vivo. Dihydroartemisinin can be regarded as an effective metabolite of artemisinin sodium, and its kinetics in dogs is a first-order rate reaction. One-compartment model characteristics, half-life i_(1/2) 10 minutes, performance volume Vd 0.36LL/kg. Artemisinin sodium and other drugs have the effect of inducing liver drug enzymes, and the first pass effect is obvious.
