目的利用人脐静脉内皮细胞(HUVEC)构建人体外结直肠癌三维血管生成模型。方法原代培养HUVEC,通过Matrigel形成三维细胞培养系统,并在该系统内共培养HUVEC和结直肠癌LoVo细胞,观察HUVEC的血管生成。结果HUVEC和结直肠癌LoVo细胞成功共培养,形成了不连续的三维类毛细血管样结构。结论所构建的结直肠癌血管生成体外三维模型能为研究直结肠癌的血管生成过程提供了一个可供选择的体外模型,也能为直结肠癌血管生成干预和抗血管生成药物的筛选提供了一个更接近体内生理状态的研究平台。
Objective To construct human three-dimensional angiogenesis model of human colorectal cancer by using human umbilical vein endothelial cells (HUVECs). Methods HUVECs were cultured in primary culture. Three-dimensional cell culture system was formed by Matrigel. HUVEC and colorectal cancer LoVo cells were co-cultured in this system to observe the angiogenesis of HUVEC. Results HUVEC and colorectal cancer LoVo cells were successfully co-cultured to form a discontinuous three-dimensional capillary-like structure. Conclusion The constructed three-dimensional model of angiogenesis of colorectal cancer in vitro can provide an alternative in vitro model for the study of angiogenesis in colorectal cancer and can also provide an interventional model for colorectal cancer angiogenesis and antiangiogenic drug screening A research platform closer to the physiological state in vivo.
