目的研究构成型光形态建成同源亚基3(COPS3)在肝癌中的表达。方法应用免疫组织化学SP法及RT-PCR法检测肝癌组织中COPS3表达情况。结果肝癌组织中COPS3蛋白表达阳性率〔46.7%(14/30)〕明显高于正常肝脏组织〔23.3%(7/30),χ2=5.654,P=0.017〕及肝硬变组织〔36.7%(11/30),χ2=4.979,P=0.026〕,差异具有统计学意义。肝癌组织中COPS3 m RNA的转录水平亦明显高于肝硬变及正常肝脏组织,差异具有统计学意义(P<0.05);在肝癌组织中,COPS3在细胞核中的表达阳性率明显高于在细胞浆中的表达阳性率(χ2=6.674,P=0.009)。结论 COPS3在肝癌组织中表达水平和定位均发生改变,可能参与了肝癌的发生和发展过程。
Objective To study the expression of COPS3 (constitutively expressed) in hepatocellular carcinoma (HCC). Methods The expression of COPS3 in hepatocellular carcinoma was detected by immunohistochemical SP method and RT-PCR. Results The positive rate of COPS3 protein in HCC tissues was 46.7% (14/30), which was significantly higher than that in normal liver tissues 〔23.3% (7/30), χ2 = 5.654, P = 0.017〕 and cirrhosis tissues 〔36.7% 11/30), χ2 = 4.979, P = 0.026], the difference was statistically significant. The transcription level of COPS3 mRNA in hepatocellular carcinoma was also significantly higher than that in cirrhosis and normal liver tissue (P <0.05). In hepatocellular carcinoma, the positive rate of COPS3 in nucleus was significantly higher than that in hepatocellular carcinoma The positive expression rate in plasma (χ2 = 6.674, P = 0.009). Conclusion The expression of COPS3 in hepatocellular carcinoma tissue and the location of both changes, may be involved in the occurrence and development of liver cancer.
