本文选用RH株弓形虫速落子细胞质、细胞膜和腹水代谢抗原,观察温度、pH值对抗原活性的影响,并分别测定糖及蛋白质含量。结果表明,三种抗原对热处理都较为稳定,但代谢抗原在加热前后的检测效果均不及其它两者。经糖含量测定显示,代谢抗原的糖含量最低。结果提示,弓形虫抗原的重要决定因子可能是糖蛋白或多糖成分。pH值试验表明,细胞质抗原的包被波以pH9.6碳酸盐缓冲液为佳,而细胞膜抗原应选用pH6.5磷酸盐缓冲液。
In this paper, Toxoplasma gondii RH strain T. gondii cytoplasm, cell membrane and ascites metabolic antigen selection, observed temperature and pH on the antigen activity, and determination of sugar and protein content. The results showed that the three antigens were relatively stable to heat treatment, but the detection effect of metabolic antigens before and after heating were lower than the other two. The determination of sugar content, the lowest content of carbohydrate antigen. The results suggest that important determinants of Toxoplasma gondii may be glycoproteins or polysaccharides. The pH test showed that the cytoplasmic antigen was coated with pH9.6 carbonate buffer, while the membrane antigen should use pH6.5 phosphate buffer.
