目的探讨β-连环蛋白(β-catenin)和富含亮氨酸的G蛋白偶联受体蛋白5(Lgr5)基因在先天性巨结肠(HD)中的表达。方法采用RT-PCR和Western blot检测30例HD患者正常肠段(有神经节细胞)和狭窄肠段(无神经节细胞)中β-catenin和Lgr5mRNA和蛋白表达,免疫组化检测β-catenin和Lgr5阳性表达情况。结果β-Catenin和Lgr5mRNA相对表达量在HD狭窄肠段中分别为17.66±1.28和15.01±1.29,低于正常肠段中的31.77±2.21和29.61±3.23(P<0.05)。β-Catenin和Lgr5相对蛋白含量在HD狭窄肠段中分别为19.51±1.57和17.35±0.87,低于正常肠段中的37.05±1.23和33.17±2.09(P<0.05)。β-Catenin和Lgr5在HD狭窄肠段中呈弱阳性或阴性表达。结论β-Catenin和Lgr5异常表达与HD发生发展相关。
Objective To investigate the expression of β-catenin and leucine-rich G protein coupled receptor 5 (Lgr5) gene in Hirschsprung’s disease. Methods The expressions of β-catenin and Lgr5 mRNA and protein in normal intestine (with ganglion cells) and narrow intestine (no ganglion cells) in 30 patients with HD were detected by RT-PCR and Western blot. The expressions of β-catenin and Lgr5 positive expression. Results The relative expression of β-Catenin and Lgr5 mRNA was 17.66 ± 1.28 and 15.01 ± 1.29 in the narrow segment of the intestine, which was lower than that in the normal segment (31.77 ± 2.21 and 29.61 ± 3.23, P <0.05). The relative protein content of β-Catenin and Lgr5 were 19.51 ± 1.57 and 17.35 ± 0.87 in the narrow segment of the HD, respectively, which was lower than that in the normal segment (37.05 ± 1.23 and 33.17 ± 2.09, P <0.05). β-Catenin and Lgr5 are weakly or negatively expressed in the narrow segment of the HD. Conclusion The abnormal expressions of β-Catenin and Lgr5 are associated with the development of HD.
