目的观察紫杉醇诱导的神经病理性大鼠脊髓背角谷氨酸转运体-1(GLT-1)的含量变化。方法雄性SD大鼠45只,随机分成正常对照组(A组),紫杉醇处理组(B组)和紫杉醇溶剂组(C组),A组不作任何处理,B组隔天注射2mg/1 000g体重的紫杉醇,C组隔天注射与B组容积相等的紫杉醇溶剂二甲基亚砜。每天检测大鼠的机械缩足阈值(MWT)。注射的第5d、7d、9d各组均处死5只大鼠,取脊髓背角组织,采用蛋白印迹方法检测各组大鼠GLT-1的表达。结果 B组大鼠的机械缩足阈值在注射第6d时显著降低(P<0.05),并且明显低于A、C组(P<0.05)。A、C两组的MWT值无变化。B组大鼠GLT-1的表达在注射第5d起明显低于A、C两组(P<0.05)。B组大鼠的GLT-1/β-actin的比值与机械缩足阈值呈显著性正相关(r=0.820,P<0.05)。结论紫杉醇诱导的神经病理性疼痛大鼠机械痛敏明显增加,脊髓背角GLT-1表达下调,GLT-1可能参与了紫杉醇致神经病理性疼痛的发生。
Objective To observe the change of glutamate transporter 1 (GLT-1) in spinal dorsal horn induced by paclitaxel in neuropathic rats. Methods 45 male SD rats were randomly divided into normal control group (A group), paclitaxel treatment group (B group) and paclitaxel solvent group (Group C). Group A received no treatment at 2 mg / kg body weight Of paclitaxel, group C was injected with paclitaxel solvent dimethylsulfoxide of volume equal to that of group B on the next day. Rats were examined daily for mechanical contracting threshold (MWT). On the 5th, 7th, and 9th day after injection, 5 rats in each group were sacrificed and the spinal dorsal horn tissues were taken. The expression of GLT-1 in each group was detected by Western blotting. Results The mechanical contracting threshold of rats in group B decreased significantly (P <0.05) on the 6th day after injection and was significantly lower than those in groups A and C (P <0.05). There was no change in the MWT values of A and C groups. The expression of GLT-1 in group B was significantly lower than that in groups A and C on the 5th day after injection (P <0.05). The ratio of GLT-1 / β-actin in group B was significantly and positively correlated with the threshold of mechanical contractions (r = 0.820, P <0.05). Conclusion The mechanical pain sensitivity of paclitaxel-induced neuropathic pain rats was significantly increased, the expression of GLT-1 in spinal dorsal horn was down-regulated, and GLT-1 might be involved in the pathogenesis of neuropathic pain induced by paclitaxel.
