为观察模拟失重对大鼠肺脏的影响,实验选用Wistar大鼠,采用尾悬吊(-30°)模拟失重模型.实验大鼠36只被分为三组:对照组、悬吊5.5天组及悬吊6.5天组.实验观察了悬吊5.5天和6.5天大鼠肺脏的形态学变化,测定了肺泡灌洗液中二棕榈酸磷脂酰胆硷(DL-2-phosphatidylcholine Dipalmitoyl,DSPC)的含量及其表面张力,以及血清中血管紧张素转化酶(Angiotensin converting enzyme,ACE)的活性等.结果表明,多数悬吊大鼠的肺脏出血,部位以右肺前叶和副叶多见,悬吊5.5天和6.5天组形态学变化相似.镜下肺毛细血管充盈、扩张,肺泡因受充盈毛细血管挤压而变小,部分毛细血管内皮细胞出现破裂,肺泡内可见红细胞.未发现肺水肿。ACE活性比对照组略低,无统计学显著性.DSPC变化不明显,但灌洗液的表面张力增加,这说明尾吊-30°模拟失重已影响到肺表面活性物质的功能.
