1目的 探讨山东省丙型肝炎病毒 (HCV)分离株 C区、NS5区的核苷酸序列及其基因型别。 2方法以 RT- nest- PCR方法从 1例临床 HCV感染者血清中分别扩增出 HCV C区 (432 bp)及 NS5区 (319bp)的基因片段 ,将其克隆于 T载体上 ,以双脱氧末端终止法自动测序并进行序列分析。 3结果 该分离株与 Gen Bank中 5 0多个 1b型分离株同源性均在 90 %以上 ,其中 C区与中国北京的 1b型株 HCU6 3376同源性最高 ,为 99.0 7% ,NS5区与日本 1b型株 HPCHUMR同源性最高 ,达 94.98% .两片段序列于 Gen Bank中申请登录号 ,C区为 AF2 13895 ,NS5区为 AF2 13893.4结论 该山东省 HCV分离株为 1b基因型 ,但其核苷酸序列仍有一定程度的变异
1 Objective To investigate the nucleotide sequences and genotypes of C and NS5 regions of hepatitis C virus (HCV) isolates in Shandong Province. 2 Methods The HCV C region (432 bp) and NS5 region (319 bp) gene fragments were amplified by RT-nest-PCR from the serums of one clinical HCV-infected patient, cloned into T vector, End-terminator method automatically sequenced and sequence analysis. 3 Results The isolates shared more than 90% homology with more than 50 isolates of genotype 1b in Gen Bank, of which the homology was highest with HCU6 3376 of type 1b strain in Beijing, China (99.0 7%). The NS5 region The homology of HPCHUMR to the Japanese 1b strain was the highest (94.98%). Two-segment sequence was submitted to Gen Bank for accession number AF2 13895 in region C and AF2 13893.4 in NS5 region. The nucleotide sequence is still a certain degree of variation
