目的:探讨bcl2mRNA在良性前列腺增生发病过程中所起的作用。方法:采用斑点杂交技术研究正常和增生前列腺组织中bcl2mRNA表达情况,并加以比较,同时采用原位杂交技术了解bcl2mRNA在增生前列腺中表达。结果:bcl2mRNA在增生前列腺组织中表达明显高于在正常前列腺组织中的表达,增生组织中平均表达值是正常组织的(1.71±0.72)倍;原位杂交显示:bcl2mRNA主要在前列腺上皮组织中表达,并呈现由基底细胞向表层上皮细胞表达逐渐减弱趋势。结论:bcl2mRNA在增生组织中高表达表明增生前列腺上皮细胞抗凋亡作用强于正常组织,提示bcl2参与前列腺细胞增殖与凋亡过程,增生前列腺中bcl2高表达可能是诱发良性前列腺增生的重要病因。
Objective: To investigate the role of bcl 2 mRNA in the pathogenesis of benign prostatic hyperplasia. Methods: Dot blot hybridization technique was used to study the expression of bcl2 mRNA in normal and hyperplastic prostatic tissues. The expression of bcl2 mRNA in proliferating prostate was also analyzed by in situ hybridization. Results: The expression of bcl2 mRNA in prostatic hyperplasia tissues was significantly higher than that in normal prostatic tissues. The average expression of bcl2 in normal tissues was (1.71 ± 0.72) times of that in normal tissues. In situ hybridization showed that bcl 2 mRNA is mainly expressed in prostate epithelial tissue, and showed the basal cells to the surface epithelial cells showed a gradual downward trend. Conclusion: The high expression of bcl 2 mRNA in hyperplastic tissues suggests that the antiproliferative effect of prostatic epithelial cells is stronger than that of normal tissues, which suggests that bcl-2 is involved in the process of prostatic cell proliferation and apoptosis.
