目的探讨混合稀土“常乐”染毒对糖尿病大鼠脂肪组织中过氧化物酶体增殖物激活受体-γ(PPAR-γ)和葡萄糖转运蛋白4(GLUT4)蛋白水平的影响。方法将40只健康清洁级Wistar雄性大鼠随机分为3组,分别为对照组(10只)、糖尿病组(18只)和混合稀土“常乐”组(12只)。糖尿病组和混合稀土“常乐”组腹腔注射50 mg/kg链脲佐菌素制备糖尿病模型;混合稀土“常乐”组采用灌胃方式染毒0.2 mg/kg常乐,对照组及糖尿病组均每日给予等剂量的生理盐水,1次/d,连续染毒1个月,然后处死取脂肪组织。采用流式细胞仪检测大鼠脂肪组织中PPAR-γ和GLUT4基因表达的阳性百分率;采用ELISA法检测大鼠脂肪组织中PPAR-γ和GLUT4蛋白的水平;采用苏木素-伊红(HE)染色观察大鼠脂肪组织的病理改变;采用免疫组织化学方法检测大鼠脂肪组织中PPAR-γ和GLUT4蛋白的阳性强度。结果与对照组比较,糖尿病模型组大鼠脂肪组织中PPAR-γ和GLUT4基因阳性百分率、蛋白水平及蛋白的阳性表达强度均显著降低,差异有统计学意义(P<0.01);混合稀土“常乐”组大鼠脂肪组织中PPAR-γ和GLUT4基因阳性百分率、蛋白水平及蛋白的阳性表达强度均略降低,但差异无统计学意义(P>0.05);与糖尿病组比较,混合稀土“常乐”组大鼠脂肪组织中PPAR-γ和GLUT4基因阳性百分率、蛋白水平及蛋白的阳性表达强度均明显升高,差异有统计学意义(P<0.01)。结论混合稀土“常乐”可上调糖尿病大鼠脂肪组织PPAR-γ和GLUT4的表达水平,从而对糖尿病大鼠机体有一定的保护作用。
Objective To investigate the effect of mixed rare earth “Changle ” on protein levels of peroxisome proliferator - activated receptor - gamma (PPAR - gamma) and glucose transporter 4 (GLUT4) in adipose tissue of diabetic rats. The positive intensity of PPAR-γ and GLUT4 protein in rat adipose tissue was detected by immunohistochemical method. Results Compared with the control group, the positive percentages of PPAR-γ and GLUT4 gene in adipose tissue of diabetic model group were significantly lower (P <0.01), and the levels of protein and protein were significantly decreased The positive percentage of PPAR-γand GLUT4 in adipose tissue of rats in “Changle” group were slightly decreased, but the difference was not statistically significant (P> 0.05).
