目的探讨大肠癌细胞中人类白细胞分化抗原133(CD133)与八聚体结合转录因子4(OCT4)的表达情况及其相关性。方法采用磁珠细胞分离技术分选出人结肠癌细胞系的CD133+细胞和CD133-细胞,检测两种细胞中OCT4的表达。分别改变CD133+和CD133-细胞中OCT4的表达,观察其对CD133的表达的影响。采用肿瘤球形成实验检验肿瘤细胞干性生物学行为的改变。结果 CD133+细胞中OCT4呈现高表达,CD133-细胞中OCT4基本无表达。CD133+细胞对5-氟尿嘧啶(5-FU)高度抵抗,并且更容易在无血清培养基中形成肿瘤球。干扰OCT4的表达可以降低CD133+细胞中CD133的表达水平,增加对5-FU的敏感性,并且减少肿瘤球形成能力。过表达OCT4增加CD133-细胞CD133的表达水平,降低对5-FU的敏感性,并且减少肿瘤球形成能力。结论 OCT4与CD133在大肠癌细胞中均有表达,OCT4对CD133有直接调控作用。
Objective To investigate the expression and correlation of human leukocyte differentiation antigen 133 (CD133) and octamer binding transcription factor 4 (OCT4) in colorectal cancer cells. Methods The CD133 + cells and CD133- cells of human colon cancer cell lines were sorted by magnetic bead separation and the expression of OCT4 in both cells was detected. OCT4 expression in CD133 + and CD133- cells was changed respectively to observe its effect on the expression of CD133. Tumor cell formation assay was used to test the change of stem cell biological behavior. Results The expression of OCT4 in CD133 + cells was high, and that of OCT4 in CD133- cells was almost no expression. CD133 + cells are highly resistant to 5-fluorouracil (5-FU) and are more likely to form tumor spheres in serum-free media. Interference with OCT4 expression can reduce CD133 expression in CD133 + cells, increase sensitivity to 5-FU, and decrease tumor spheroid formation. Overexpression of OCT4 increases the expression level of CD133-CD133, decreases the sensitivity to 5-FU, and reduces the tumor’s ability to form spheres. Conclusions Both OCT4 and CD133 are expressed in colorectal cancer cells. OCT4 has a direct regulatory effect on CD133.
