目的观察染料木素对实验性去卵巢大鼠诱发的骨质疏松症模型的治疗作用。方法SD雌性大鼠行去卵巢术后,随机分为正常对照组,模型组,骨疏康(GSK)组,尼尔雌醇组,染料木素大、中和小剂量组,每组10只。各组动物给予盐酸四环素25mg·kg-1连续3d,同时给予治疗用药,剂量为GSK5g·kg-1,尼尔雌醇为1.5mg·kg-1,染料木素分别为18,9,4.5mg·kg-1,以上给药除尼尔雌醇每周1次外其他各组均为每日1次。给药后12周后进行相关指标的测定。结果①染料木素4.5,9mg·kg-1连续给药12周可使其股骨干重、灰重、骨钙及股骨和腰椎骨密度显著增加,与模型组比较具有显著差异(P<0.05或P<0.01);②染料木素4.5,9mg·kg-1连续给药12周可显著改善模型大鼠的股骨及椎骨最大载荷、最大桡度、弹性载荷、弹性桡度(P<0.05或P<0.01);③染料木素低、中剂量用药12周后可明显增加模型大鼠骨小梁面积百分比(BV/TV%)、骨小梁厚度(Tb.Th)和骨小梁数目(Tb.N),减小骨小梁间隔(Tb.Sp)(P<0.05或P<0.01);④染料木素低、中剂量给药12周后,可使模型大鼠的血清ALP水平下降,尿Ca/Cr、Pyd/Cr水平降低(P<0.05或P<0.01)。结论染料木素对模型动物的骨密度、骨生物力学及相关生化指标都具有改善作用,具有较好的抗骨质疏松作用。
Objective To observe the therapeutic effect of genistein on experimental ovariectomized rat model of osteoporosis. Methods SD female rats were randomly divided into normal control group, model group, Gushukang (GSK) group, nilestriol group, genistein large and medium dose group, 10 rats in each group . Each group of animals was given tetracycline hydrochloride at 25mg · kg-1 for 3d consecutive days, while the treatment was given at the dose of GSK5g · kg-1, nylestriol 1.5mg · kg-1, genistein 18,9 and 4.5mg · Kg-1, the above administration in addition to nilestriol once a week outside the other groups are daily 1 time. After 12 weeks after administration, the related indexes were measured. Results ① Genistein at 4 and 9 mg · kg-1 administration for 12 weeks significantly increased the femoral shaft weight, weight, bone calcium, femur and lumbar BMD, with significant difference compared with model group (P <0.05 or P <0.01). ② Genistein 4.5 and 9 mg · kg-1 administration for 12 weeks significantly improved the maximal, maximal radial, elastic and elastic radii of the femur and vertebra (P <0.05 or P <0.01). (3) BV and TV%, trabecular thickness (Tb.Th) and trabecular number (Tb (P <0.05 or P <0.01). (4) After 12 weeks of administration of genistein, the level of ALP in the model rats decreased, Urinary Ca / Cr, Pyd / Cr levels decreased (P <0.05 or P <0.01). Conclusion Genistein can improve bone mineral density, bone biomechanics and related biochemical parameters in model animals, and has good anti-osteoporosis effect.
