我们对苜蓿和百脉根的原生质体融合及培养进行了研究。用5mM碘乙酰胺处理百脉根原生质体以抑制其细胞分裂,使用在培养早期分裂但未形成细胞群的苜蓿原生质体进行融合,以期望仅有异核的原生质体可以形成愈伤组织。但是,通过聚乙二醇(PEG)处理的原生质体融合仅获得了两块表现有与双亲不同特征的愈伤组织,而多数愈伤组织只表现了百脉根的特征。根据同功酶分析和染色体观察,确认有一块愈伤组织是体细胞杂种,但这块愈伤组织还没有再生出杂种植株。
We studied the protoplast fusion and culture of alfalfa and Lotus corniculatus. Xenograft protoplasts were treated with 5 mM iodoacetamide to inhibit their cell division and fused using alfalfa protoplasts that were cultured in early stages of division but not in cell population in the hope that only heteronucleated protoplasts could form callus. However, only two calluses exhibiting different characteristics from their parents were obtained by protoplast fusion with polyethylene glycol (PEG) treatment, whereas most of the calluses showed only the characteristics of Lotus root. According to isoenzyme analysis and chromosome observation, one piece of callus was confirmed to be a somatic hybrid, but the callus had not regenerated a hybrid plant yet.
