目的通过BN大鼠皮肤贴敷和皮内注射试验染毒具有致敏性的外源性化学物质己基肉桂醛,观察致敏特点,同时以豚鼠作为对照观察动物,探讨BN大鼠作为化学品过敏试验动物模型的可能性。方法将BN大鼠和白色豚鼠分别随机分为2组:对照组和己基肉桂醛组,每组10只。将己基肉桂醛组实验动物于试验的第1天分别进行皮肤贴敷试验和皮内注射试验,皮肤贴敷试验于首次贴敷后28 d进行激发试验,皮内注射试验于皮内注射后21 d进行激发试验;对照组给予相同体积的生理盐水注射液。激发后观察每只动物过敏反应情况,根据过敏反应症状评分,计算致敏率,对致敏强度进行分级。经腹主动脉取血,分离血清,竞争ELISA法检测血清Ig E含量。结果皮肤贴敷试验中染毒己基肉桂醛后,BN大鼠和豚鼠致敏率分别为60%和50%;BN大鼠血清Ig E含量与对照组相比差异有统计学意义(P<0.05),豚鼠Ig E含量与对照组相比差异无统计学意义(P>0.05)。皮内注射试验中染毒己基肉桂醛后,BN大鼠和豚鼠致敏率分别为70%和80%;BN大鼠和豚鼠己基肉桂醛组血清Ig E含量与各自的对照组相比,差异有统计学意义(P<0.05)。结论在皮肤贴敷和皮内注射过敏反应中,BN大鼠已基肉桂醛组呈阳性反应,染毒后的局部反应特点强于豚鼠,因此,BN大鼠作为化学品过敏试验的动物模型具备可能性。
OBJECTIVE To investigate the allergenicity of hexyl cinnamic aldehyde, an allergenic exogenous chemical substance, by applying skin and intradermal injection on BN rats and observe the sensitization characteristics. At the same time, guinea pigs were used as controls to observe the animals as BN rats as chemical allergy Possibility of testing animal models. Methods BN rats and white guinea pigs were randomly divided into 2 groups: control group and hexyl cinnamic aldehyde group, 10 rats in each group. The cinnamic aldehyde group of hexadecyl aldehyde test animals were tested on the first day of the skin adhesion test and intradermal injection test, skin adhesion test 28 days after the first application of the excitation test, intradermal injection test after intradermal injection of 21 d for excitation test; the control group was given the same volume of saline injection. After the challenge, each animal was observed for the allergic reaction. According to the symptom score of the allergic reaction, the sensitization rate was calculated and the sensitization intensity was graded. Serum was collected from the abdominal aorta and serum was separated. IgE level in the serum was detected by competitive ELISA. Results The sensitivities of BN rats and guinea pigs were 60% and 50% respectively after being inoculated with hexylcinnamaldehyde in skin patch test. The serum IgE levels in BN rats were significantly different from those in control group (P <0.05 ), There was no significant difference in IgE content between guinea pigs and control group (P> 0.05). The sensitivities of BN rats and guinea pigs were 70% and 80%, respectively, after inoculation with hexylcinnamaldehyde in the intradermal injection test. The serum IgE levels in BN rats and guinea pig hexylcinnamaldehyde groups were significantly lower than those in the control group There was statistical significance (P <0.05). Conclusion In the dermal application and intradermal injection of anaphylactic reaction, the BN rat has a positive reaction with a base cinnamaldehyde group, and the local reaction characteristic after exposure is stronger than that of a guinea pig. Therefore, the BN rat is an animal model of chemical allergy test possibility.
