[目的]探讨人骨髓核分化抗原(MNDA)对人骨肉瘤Saos-2细胞增殖及凋亡的影响。[方法]逆转录聚合酶链反应(RT-PCR)法定量测定Saos-2细胞中MNDA的mRNA表达差异。将MNDA基因转染Saos-2细胞,应用MTT法测定转染后的肿瘤细胞增殖,流式细胞仪定量检测细胞凋亡程度,没有转染质粒的和转染空质粒的做为对照组。[结果]MNDA的mRNA在骨肉瘤Saos-2细胞中表达明显降低。转染MNDA后人骨肉瘤Saos-2细胞的凋亡程度高于对照组,增殖率低于对照组。[结论]转染MNDA可诱导人骨肉瘤Saos-2细胞发生凋亡,抑制骨肉瘤细胞的增殖率。
[Objective] To investigate the effect of human bone marrow nuclear antigen (MNDA) on the proliferation and apoptosis of human osteosarcoma Saos-2 cells. [Method] The mRNA expression of MNDA in Saos-2 cells was quantitatively determined by reverse transcription-polymerase chain reaction (RT-PCR). The MNDA gene was transfected into Saos-2 cells. MTT assay was used to determine the proliferation of tumor cells. Flow cytometry was used to detect the degree of apoptosis. The transfected cells were transfected with empty plasmid. [Results] The mRNA expression of MNDA was significantly decreased in osteosarcoma Saos-2 cells. The apoptosis of human osteosarcoma Saos-2 cells after transfection with MNDA was higher than that of the control group, and the proliferation rate was lower than that of the control group. [Conclusion] Transfection of MNDA can induce the apoptosis of human osteosarcoma Saos-2 cells and inhibit the proliferation rate of osteosarcoma cells.
