目的建立雷公连与穿心藤的薄层色谱、紫外-可见光谱鉴别方法。方法利用紫外分光光度计测定雷公连、穿心藤的水、无水乙醇、氯仿和石油醚(60~90℃)4种溶剂的浸泡液的紫外-可见吸收图谱,采用紫外-可见谱线组的吸收峰数及峰位置对其结构特征进行分析鉴别;采用水、正丁醇、石油醚(60~90℃)依次超声提取雷公连、穿心藤,以石油醚-丙酮(8∶2)、石油醚-丙酮(9∶2)、石油醚-氯仿(3∶2)为展开剂,用硅胶G预制薄层板,以10%的硫酸乙醇液为显色剂,进行薄层色谱分析。结果雷公连、穿心藤的紫外-可见光谱图的最大吸收峰数及峰位置有差异,其薄层色谱主斑点位置及颜色有明显差异,且集中、分离度好。结论所建立的方法快速、简便、可靠、灵敏、重现性好、专属性强,可为雷公连、穿心藤的药材鉴别提供参考。
Objective To establish a TLC and UV-Vis spectrophotometric method for the determination of Thunb and thunb. Methods Ultraviolet-visible absorption spectra of the soaking solution of four kinds of solvents, including Thunder, Trifolium repens, absolute ethanol, chloroform and petroleum ether (60-90 ℃), were determined by ultraviolet spectrophotometer. The UV- (8: 2) were extracted by sonication with water, n-butanol and petroleum ether (60 ~ 90 ℃) , Petroleum ether - acetone (9: 2) and petroleum ether - chloroform (3: 2) as developing solvent. The thin layer was chromatographed on silica gel G and TLC with 10% sulfuric acid solution. Results The maximum absorption peaks and the peak positions of UV-Vis spectra of Thunder and Kadsura were different. The location and color of the main spots in TLC were significantly different, and the concentration and resolution were good. Conclusion The established method is rapid, simple, reliable, sensitive, reproducible and specific. It can provide a reference for identification of Radix Rehmanniae and Cordyceps.
