目的研究不同剂量的γ射线对人胰腺癌细胞凋亡及bcl2,bax基因表达的影响。方法利用不同剂量的γ射线对体外培养的胰腺癌Panc1细胞进行照射,采用PI和AnnexinⅤPI法定量检测细胞凋亡,流式细胞仪检测照射后胰腺癌细胞Panc1的Bcl2、Bax基因表达水平。结果胰腺癌panc1细胞凋亡百分率在一定剂量范围内(≤15Gy)随着照射剂量的提高而增大,在一定时间范围内(≤24h),随着时间的延长而增大。照射组细胞凋亡相关基因Bcl2表达较对照组明显降低,两组相比差异有统计学意义(P<005);照射组bax基因的表达较对照组明显升高,两组相比差异有统计学意义(P<005)。结论γ射线诱导胰腺癌panc1细胞凋亡具有剂量依赖性和时间相关性,bcl2和bax在胰腺癌细胞凋亡调节过程中起着重要作用,不同剂量γ射线照射胰腺癌细胞时Bcl2和Bax表达水平也不相同,通过降低bcl2的表达及提高bax表达来诱导胰腺癌细胞发生凋亡有可能是γ射线杀伤肿瘤细胞的机制之一。
Objective To study the effects of different doses of γ-rays on the apoptosis of human pancreatic cancer cells and the expression of bcl-2 and bax genes. Methods Panc1 cells cultured in vitro were irradiated with different doses of γ-rays. The apoptosis rate of Panc1 cells was detected by PI and AnnexinⅤPI.
