目的:探讨姜黄素对大鼠HSC-T6表达PDGF-BB、PDGFRβ、ERK1的影响。方法:采用MTT法观察不同质量浓度姜黄素对HSC细胞增殖的影响,并计算其抑制率,确定姜黄素最佳等比浓度,设对照组、TGF-β1刺激组和姜黄素各质量浓度干预组。采用免疫组化、聚合酶链式反应(RT-PCR)半定量法探讨姜黄素对HSC-T6PDGF-BB、PDGFRβ、ERK1表达的影响。结果:正常培养的HSC-T6均有PDGF-BB、PDGFRβ、ERK1表达,经姜黄素不同质量浓度干预后,PDGF-BB、PDGFRβ、ERK1表达降低,且有一定的剂量依赖性。结论:姜黄素能抑制HSC-T6PDGF-BB、PDGFRβ、ERK1的表达,这可能是其抗肝纤维化的作用机理。
Objective: To investigate the effects of curcumin on the expression of PDGF-BB, PDGFRβ and ERK1 in rat HSC-T6. METHODS: MTT assay was used to observe the effects of different concentrations of curcumin on the proliferation of HSC cells, and the inhibitory rate was calculated to determine the best concentration of curcumin. The control group, TGF-β1 stimulation group and curcumin concentration intervention group were used. . The effects of curcumin on the expression of HSC-T6PDGF-BB, PDGFRβ, and ERK1 were investigated by immunohistochemistry and polymerase chain reaction (RT-PCR). Results: The expressions of PDGF-BB, PDGFRβ and ERK1 were detected in normal cultured HSC-T6. The expression of PDGF-BB, PDGFRβ, and ERK1 decreased after curcumin treatment with different concentrations, and there was a dose-dependent manner. Conclusion: Curcumin can inhibit the expression of HSC-T6PDGF-BB, PDGFRβ, ERK1, which may be the mechanism of anti-hepatic fibrosis.
