目的建立HPLC测定瑞替加滨含量的方法。方法色谱柱为Agilent ZORBAX SB-C18(150 mm×4.6 mm,3.5μm),流动相为庚烷磺酸钠溶液-乙腈,采用梯度洗脱,流速为1.0 mL.min 1,检测波长为250 nm,温度为30℃。结果瑞替加滨在0.204~122.28μg.mL 1内,峰面积与浓度呈良好的线性关系(r=0.999 9),平均回收率99.9%,RSD=0.46%。结论本法准确、重复性好,能与其同分异构体较好分离,适用于瑞替加滨的含量测定。
Objective To establish a HPLC method for the determination of retigabine. Methods The column was Agilent ZORBAX SB-C18 (150 mm × 4.6 mm, 3.5 μm). The mobile phase consisted of heptane sulfonate solution-acetonitrile. The gradient elution was performed at a flow rate of 1.0 mL · min 1 and the detection wavelength was 250 nm , The temperature is 30 ℃. Results Retigabine showed a good linearity (r = 0.999 9) with an average recovery of 99.9% and a RSD of 0.46% in the range of 0.204 to 122.28 μg · mL 1. Conclusion This method is accurate, reproducible and can be separated from its isomers well and is suitable for the determination of retigabine.
