细胞周期抑制蛋白p27~(kip1)和周期蛋白表达与大肠癌发生预后(英文)

目的探讨细胞周期调控抑制因子p27kip1蛋白和周期蛋白E(cyclinE)在大肠腺癌发生发展中的作用。方法采用免疫组化技术SP法,检测p27kip1和cyclinE蛋白在37例大肠腺癌和26例正常大肠黏膜组织中的表达。结果p27kip1蛋白阳性表达率在大肠腺癌组织中为45.9%,显著低于正常大肠黏膜组织(χ2=4.590,P<0.05),并与大肠腺癌组织分化程度和淋巴结转移均有关(χ2=9.653,χ2=6.838,P<0.05);cyclinE蛋白阳性表达率在大肠腺癌组织中为64.9%,显著高于正常大肠黏膜组织(χ2=4.285,P<0.05),并与大肠腺癌组织分化程度和淋巴结转移均有关(χ2=8.397,χ2=5.261,P<0.05);p27kip1蛋白阳性表达组cyclinE蛋白阳性表达率低于p27kip1蛋白阴性表达组,但两者无相关(r=-0.1133,P>0.05)。结论p27kip1蛋白的低表达和/或cyclinE蛋白的高表达在大肠腺癌发生中发挥重要作用,并可能有助于预测其恶性程度及淋巴结转移,可能对大肠腺癌的治疗和生活质量的改善有指导作用。 Objective To investigate the role of cell cycle regulatory inhibitor p27kip1 and cyclinE in the development of colorectal adenocarcinoma. Methods Immunohistochemical SP method was used to detect the expressions of p27kip1 and cyclinE in 37 cases of colorectal adenocarcinoma and 26 cases of normal colorectal mucosa. Results The positive expression rate of p27kip1 protein was 45.9% in colorectal adenocarcinoma, which was significantly lower than that in normal colorectal mucosa (χ2 = 4.590, P <0.05), and was correlated with the degree of differentiation and lymph node metastasis in colorectal adenocarcinoma (χ2 = 9.653 , χ2 = 6.838, P <0.05). The positive rate of cyclinE protein in colorectal adenocarcinoma was 64.9%, which was significantly higher than that in normal colorectal mucosa (χ2 = 4.285, P <0.05) (Χ2 = 8.397, χ2 = 5.261, P <0.05). The positive rate of cyclinE protein in p27kip1 positive group was lower than that in negative p27kip1 group (χ2 = 8.397, P> 0.05). Conclusions The low expression of p27kip1 protein and / or high expression of cyclinE protein play an important role in the development of colorectal adenocarcinoma and may be helpful to predict the malignancy and lymph node metastasis, and may improve the treatment of colorectal adenocarcinoma and the quality of life Guidance.
