本文研究了生物液中甲氨喋呤(MTX)的测定方法。10只家兔,体重2.5~3kg,剂量为40~30 mg/kg;恶性肿瘤患者10人,年龄8~53岁,肝肾功能正常,每次给以1~4g的MTX。注入2~4小时后,以甲酰四氢叶酸钙(CF)救援,CF(9~12mg)肌注,每6小时一次,共12次。兔和病人生物液中MTX经提取后,在UV-210紫外分光光度计306nm处测定,MTX的标准曲线方程Y=0.0456X+0.000092;血浆MTX校正曲线方程Y=0.0441 X+0.0389;尿中MTX校正曲线方程Y=0.0469+0.0417;用本法测得MTX的回收率约95%。 为了HD-MTX-CFR的安全应用,我们研究了MTX的血浆水平和尿中MTX的排泄。
This article studies the determination of methotrexate (MTX) in biological fluids. 10 rabbits weighing 2.5-3 kg at a dose of 40-30 mg / kg; 10 patients with malignant tumors aged 8-53 years with normal liver and kidney function, each receiving 1 to 4 g of MTX. After 2 to 4 hours of infusion, calcium leucovorin (CF) rescue, CF (9 ~ 12mg) intramuscular injection once every 6 hours, a total of 12 times. For safety applications of HD-MTX-CFR, we investigated the plasma levels of MTX and the excretion of MTX in the urine.
