目的研究抗菌蛋白血小板型磷脂酶A2(PLA2)、杀菌/渗透增强蛋白(BPI)和β-防御素-1mRNA在正常大鼠生殖系统内的存在情况,以了解生殖系统的天然抗菌机制的状况。方法从大鼠生殖系统不同器官和组织的匀浆中提取总RNA,合成第1链cDNA,利用PCR扩增大鼠血小板型PLA2、BPI和β-防御素-1的DNA片段,在凝胶电泳的胶上观察PCR产物条带。结果在大鼠的7种生殖器官和组织中,睾丸、附睾和卵巢均能检测到血小板型PLA2、BPI和β-防御素-1的mRNA,其中以卵巢中的血小板型PLA2mRNA、睾丸中的BPImRNA和附睾中的β-防御素-1mRNA含量尤为丰富;另外,在前列腺、子宫、阴道中也能检测到β-防御素-1和血小板型PLA2的mRNA。结论血小板型PLA2、BPI和β-防御素-1在大鼠雄性和雌性生殖系统中的多种器官和组织存在,提示天然免疫的重要成分抗菌蛋白在防御生殖系感染方面起了重要的作用。
Objective To study the existence of antimicrobial protein PLA2, bactericidal / osmotic enhanced protein (BPI) and β-defensin-1 mRNA in normal rat reproductive system to understand the natural antibacterial mechanism of reproductive system. Methods Total RNA was extracted from the homogenates of different organs and tissues of the rat reproductive system to synthesize the first strand cDNA. The DNA fragments of rat platelet PLA2, BPI and β-defensin-1 were amplified by PCR and analyzed by gel electrophoresis The bands of the PCR product were observed. Results The mRNAs of platelet PLA2, BPI and β-defensin-1 were detected in testis, epididymis and ovary in 7 kinds of reproductive organs and tissues of rats, including platelet PLA2 mRNA in ovary, BPI mRNA in testis And epididymal β-defensin-1 mRNA content is particularly rich; In addition, in the prostate, uterus, vagina also detect beta-defensin-1 and platelet-type PLA2 mRNA. Conclusion The existence of platelet-type PLA2, BPI and β-defensin-1 in various organs and tissues of male and female reproductive system suggests that antimicrobial protein, an important component of innate immunity, plays an important role in defense against germ-line infection.
