突变的ras21蛋白常发生单个氨基酸置换而成为肿瘤特异性抗原。作者对胰腺癌、结肠癌患者是否存在着对突变ras癌基因蛋白特异的T细胞或抗体进行了临床研究。首先采用ELISA法检测了160例结肠癌患者及40例正常对照者血清中是否存在抗rasD12抗体。抗体检测结果显示:31.8%病人血清可检出抗rasD12蛋白的IgA抗体,而正常对照组仅1例阳性,占2.5%。结肠癌病人血清中抗rasD12抗体明显高于正常组,说明突变ras癌基因蛋白可诱导机体产生相应抗体反应。进一步采用3H-TdR掺入法检测16例胰腺癌、28例结肠癌及11例正常人外周血中是否存在对突变ras蛋白和多肽片段致敏的T细胞。结果显示,43.7%胰腺癌病人T细胞对突变rasD12多肽呈阳性反应,而11例正常人则完全阴性。对4例对突变rasD12多肽呈阳性反应的胰腺癌患者进一步检测其T细胞能否同时对突变rasD12蛋白有特异识别,结果3例患者T细胞能同时对突变rasD12多肽和蛋白呈阳性反应。11例正常对照同样对突变rasD12蛋白无反应。在结肠癌病人中,其T细胞对rasD12多肽反应者为8.0%(2/25)。实验结果表明,突变ras蛋白的突变片段具免疫?
The mutated ras21 protein often undergoes a single amino acid substitution and becomes a tumor-specific antigen. The authors conducted a clinical study of the presence or absence of ras oncogene-specific T cells or antibodies in pancreatic and colon cancer patients. Firstly, ELISA was used to detect the presence of anti-rasD12 antibodies in the serum of 160 patients with colon cancer and 40 normal controls. The results of antibody detection showed that 31.8% of patients’ serum could detect IgA antibodies against rasD12 protein, while only 1 case of normal control group was positive, accounting for 2.5%. The anti-rasD12 antibody in the serum of patients with colon cancer was significantly higher than that in the normal group, indicating that the mutant ras oncogene protein can induce the body to produce the corresponding antibody response. Further 3H-TdR incorporation assay was used to detect the presence of T cells sensitized to mutant ras protein and polypeptide fragments in 16 cases of pancreatic cancer, 28 cases of colon cancer, and 11 cases of normal human peripheral blood. The results showed that 43.7% of pancreatic cancer patient T cells were positive for the mutant rasD12 polypeptide, whereas 11 normal people were completely negative. In addition, four cases of pancreatic cancer positive to mutant rasD12 polypeptides were further tested for their ability to specifically recognize the mutant rasD12 protein simultaneously. As a result, T cells in three cases were positive for the mutant rasD12 polypeptide and protein. Eleven normal controls also did not respond to the mutant rasD12 protein. In patients with colon cancer, their T-cell response to rasD12 polypeptide was 8.0% (2/25). The experimental results show that the mutant fragment of the mutant ras protein is immune?
