目的:探讨脾虚痰浊证巴马小型猪心肌组织葡萄糖代谢相关分子的变化特点。方法:普通级雄性巴马小型猪10只随机分为正常组及脾虚痰浊组,每组5头。采用疲劳过度联合高脂单笼饲养建立小型猪脾虚痰浊证模型。采用实时荧光定量PCR法检测心肌组织葡萄糖转运体4(glucose transporter 4,GLUT4)、磷酸果糖激酶1(Phosphofructokinase-1,PFK-1)、丙酮酸激酶(Pyruvate kinase2,PKM2)和己糖激酶(Hexokinase,HK)mRNA表达,应用Western blotting法检测心肌组织GLUT4、PFK1、PKM2和HK蛋白表达水平。结果:与正常组小型猪相比,脾虚痰浊组小型猪心肌组织GLUT4、PKM2、PFK1、HK mRNA表达水平显著下降(P<0.01),GLUT4、PKM2蛋白表达水平显著下降(P<0.05)。PFK1、HK蛋白表达水显著下降P<0.01)。结论:脾虚痰浊证小型猪心肌存在葡萄糖代谢异常,与葡萄糖转运及代谢相关分子表达呈下降趋势。
Objective: To investigate the changes of glucose metabolism-related molecules in the myocardium of Bama miniature pig with phlegm-phlegm syndrome of spleen deficiency. Methods: Ten normal male Bama miniature pigs were randomly divided into normal group and spleen deficiency phlegm group (n = 5). A model of phlegm and phlegm syndrome with spleen deficiency in spleen of piglets was established by over-fat combined with high-fat single cage. The expression of GLUT4, phosphofructokinase-1 (PFK-1), pyruvate kinase 2 (PKM2) and hexokinase HK) mRNA expression. The protein expression of GLUT4, PFK1, PKM2 and HK in myocardium was detected by Western blotting. Results: Compared with the normal group, the expression of GLUT4, PKM2, PFK1 and HK mRNA in the Jianpi phlegm turbidity mini group decreased significantly (P <0.01) and the protein expression of GLUT4 and PKM2 decreased significantly (P <0.05). PFK1, HK protein expression decreased significantly P <0.01). Conclusion: The spleen deficiency phlegm turbidity mini-pig myocardial glucose metabolism, glucose transport and metabolism-related molecules showed a downward trend.
