[目的]研究脾虚型小鼠小肠上皮细胞蛋白质组特征及白参的调节作用。[方法]昆明种小鼠60只,利血平法复制动物模型,随机分为正常、脾虚模型和白参汤剂组,双向凝胶电泳技术分离蛋白质,改进的考染法染色,以PDQuest软件分析图谱,基质辅助激光解吸电离飞行时间质谱(MALDI-TOF-MS)进行质谱分析。[结果]鉴定出的33个蛋白质点中有27个表达明显减弱,6个表达增强。经白参汤剂治疗后,其表达多数得以回调。各蛋白质的功能类型几乎涉及小肠细胞各主要功能。[结论]脾虚型小鼠小肠上皮细胞的蛋白质组特征主要表现为蛋白质表达呈双向变化,以下调为主。表达变化的蛋白质涉及细胞各部位,白参汤剂对小肠上皮细胞蛋白质组的调节作用是白参健脾的重要机制之一。
[Objective] To study the proteomic characteristics of small intestinal epithelial cells and the regulation of white ginseng in spleen deficiency mice. [Methods] Sixty Kunming mice were randomly divided into normal, spleen-deficiency model and white ginseng decoction group. The two-dimensional gel electrophoresis technique was used to separate proteins, and the improved dyeing method was used for PDQuest software. Analytical spectra were analyzed by matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF-MS) for mass spectrometry analysis. [Results] 27 out of the 33 protein spots identified were significantly decreased and 6 expressions were enhanced. After treatment with white ginseng decoction, most of its expression has been corrected. The functional type of each protein involves almost every major function of the small intestine cells. [Conclusion] The proteomic characteristics of small intestine epithelial cells of spleen deficiency mice mainly showed that the protein expression showed bidirectional changes, and the down-regulation was mainly. Proteins that express changes are involved in various parts of the cell. The function of the white ginseng decoction on the proteome of the small intestine epithelial cells is one of the important mechanisms of the white ginseng and the spleen.
