目的研究硫氧还蛋白结合蛋白(TXNIP)、前列腺跨膜上皮抗原2(STEAP2)、转录因子(GATA)家族中成员GATA3与糖尿病伴乳腺癌的关系,并分析患者术前及术后的血清氧化应激指标变化。方法检测45例糖尿病伴乳腺癌患者癌组织及对应癌旁组织中TXNIP、STAMP2、GAT.A3 mRNA表达量、蛋白水平及蛋白阳性表达情况,分析其临床病理特征,以及血清氧化应激指标的变化。结果乳腺癌组织中TXNIP mRNA、STAMP2 mRNA、GATA3 mRNA的相对表达量均下降(P<0.05)。TXNIP、STAMP2、GATA3蛋白在乳腺癌组织中的表达低于癌旁正常组织(P<0.05),且3个指标的表达阳性率为60%、66.7%、73.3%,均高于对应癌旁正常组织中的TXNIP蛋白表达阳性率(2.2%)(P<0.05)。乳腺癌组织中TXNIP、STAMP2、GATA3蛋白表达阳性率均与分化程度、淋巴结转移与临床分级相关(P<0.05)。患者术后血清MDA、髓过氧化物酶(MPO)均呈现先上升后下降变化趋势,SOD及总抗氧化力(TAC)呈现先下降后上升的变化趋势,术后6 d为转折点。结论 TXNIP、STAMP2、GATA3及血清氧化应激指标可能成为糖尿病伴乳腺癌患者诊断及治疗的参考指标。
Objective To investigate the relationship between GATA3 and diabetes mellitus associated with breast cancer in patients with TXNIP, STEAP2 and GATA. Stress indicators change. Methods The expressions of TXNIP, STAMP2 and GAT.A3 mRNA in 45 cases of diabetic patients with breast cancer and their corresponding paracancerous tissues were detected by immunohistochemistry. The clinicopathological characteristics and the changes of serum oxidative stress index . Results The relative expression of TXNIP mRNA, STAMP2 mRNA and GATA3 mRNA in breast cancer tissues decreased (P <0.05). The expression of TXNIP, STAMP2 and GATA3 in breast cancer tissue was lower than that in adjacent normal tissues (P <0.05), and the positive rates of three markers were 60%, 66.7% and 73.3% The positive rate of TXNIP protein expression in tissues (2.2%) (P <0.05). The positive rates of TXNIP, STAMP2 and GATA3 in breast cancer tissues were correlated with the degree of differentiation, lymph node metastasis and clinical grade (P <0.05). The levels of serum MDA and myeloperoxidase (MPO) showed the trend of first rising and then decreasing, the activity of SOD and total antioxidant capacity (TAC) first decreasing and then increasing, the turning point was 6 days after operation. Conclusion TXNIP, STAMP2, GATA3 and serum oxidative stress indicators may become a reference index for the diagnosis and treatment of patients with diabetes mellitus and breast cancer.
