金匮肾气丸对微波辐射性生殖功能障碍小鼠生精细胞膜流动性的影响

目的:探讨微波辐射对小鼠生精细胞膜流动性的影响及金匮肾气丸治疗机制。方法:雄性BALB/c小鼠50只,随机分为5组,每组10只。平行对照组不做任何处理;辐射对照组、金匮肾气丸1组、金匮肾气丸2组、金匮肾气丸3组均暴露于频率为2 450MHz连续微波中,功率密度为10m W/cm2,辐射18d,每天辐射1h;辐射对照组用0.9%氯化钠溶液分别灌胃,每天1次,每次0.1m L;金匮肾气丸组用金匮肾气丸(0.1、0.2、0.4g/kg体质量)分别灌胃,每天1次。组合酶法分离各组小鼠生殖细胞,荧光偏振法及激光扫描共聚焦显微镜(LSCM)检测膜微黏度(η)、荧光偏振度(P)。结果:微波辐射后,P及η显著升高(P<0.05);金匮肾气丸治疗后,P及η皆有显著改善(P<0.05),金匮肾气丸1组、2组、3组均接近正常,3组之间无显著差异。结论:微波辐射可以使小鼠生精细胞膜流动性下降,而金匮肾气丸的治疗机制是通过增加小鼠精子膜的流动性而起到治疗作用。 Objective: To investigate the effect of microwave radiation on the fluidity of spermatogenic cell membrane in mice and the therapeutic mechanism of Jingui Shenqi Pill. Methods: 50 male BALB / c mice were randomly divided into 5 groups with 10 in each group. Radiation control group, Jinguishenqiwan group 1, Jinguishenqiwan group 2 and Jinguishenqiwan group were all exposed to continuous microwave with a frequency of 2 450MHz and the power density was 10m W / cm2, radiation 18d, radiation 1h daily; radiation control group were treated with 0.9% sodium chloride solution were administered once a day, each 0.1m L; Jingui Shenqi pill group with Jingui Shenqi pill (0.1, 0.2, 0.4g / kg body weight) were administered orally, once daily. Bacterial germ cells were isolated by combinatorial enzymatic method. The microviscosity (η) and fluorescence polarization (P) of the membrane were detected by fluorescence polarization and laser scanning confocal microscopy (LSCM). Results: After microwave irradiation, P and η were significantly increased (P <0.05). After treatment with Jinkuishenqi Pill, both P and η were significantly improved (P <0.05). Jingui Shenqi Pills group 1, 2, Three groups were close to normal, no significant difference between the three groups. CONCLUSION: Microwave radiation can decrease the fluidity of spermatogenic cells in mice, while the therapeutic mechanism of Jingui Shenqi Pills plays a therapeutic role by increasing the fluidity of sperm membranes in mice.
