[目的]探讨太白楤木对肝纤维化模型大鼠Ⅰ型胶原纤维(Col-Ⅰ)、Ⅲ型胶原纤维(Col-Ⅲ)、α-平滑肌肌动蛋白(α-SMA)表达的干预作用。[方法]雄性SD大鼠60只,随机分为阴性对照组、模型组、太白楤木大、中、小剂量组和秋水仙碱组,采用40%CCl4橄榄油腹腔注射诱导肝纤维化模型,2次/周,连续8周。采用苏木精-伊红及Mas-son染色观察肝组织病理学改变;RT-PCR检测Col-Ⅰ、Col-ⅢmRNA表达水平;Western-blot检测α-SMA的表达水平。[结果]与模型组相比,太白楤木大、中、小剂量组及秋水仙碱组肝细胞变性坏死减轻,纤维条索减少,且秋水仙碱组、太白楤木小、中、大剂量组呈逐渐减轻趋势。与模型组相比,太白楤木大、中、小剂量组及秋水仙碱组Col-Ⅰ、III mRNA表达水平均减低(均P<0.05);与秋水仙碱组比较,太白楤木大剂量组Col-Ⅰ、III mRNA表达减低(均P<0.05)。太白楤木大、中、小剂量组和秋水仙碱与模型组相比,a-SMA表达不同程度减低(均P<0.05),与秋水仙碱组相比,太白楤木大、中剂量组a-SMA表达均减低(均P<0.05)。[结论]太白楤木可通过抑制肝组织中胶原纤维的增生和肝星状细胞的活化,从而减轻肝纤维化。
[Objective] To investigate the effect of Taibai on the expression of type Ⅰ collagen fibers (Col-Ⅰ), type Ⅲ collagen fibers (Col-Ⅲ) and α-smooth muscle actin (α-SMA) in rats with liver fibrosis. [Methods] Sixty male Sprague-Dawley rats were randomly divided into the negative control group, the model group, the Taibai alburnum large, medium and small dose groups and the colchicine group. The model rats were induced by intraperitoneal injection of 40% CCl4 olive oil, 2 times / week for 8 weeks. The hepatic histopathological changes were observed by hematoxylin-eosin and Mas-son staining. The expression of Col-Ⅰ and Col-Ⅲ mRNA was detected by RT-PCR. The expression of α-SMA was detected by Western-blot. [Result] Compared with the model group, the hepatocyte degeneration and necrosis, the reduction of fiber cords in the large, middle and small dose groups and the colchicine groups of the Taibai alder, and the small, medium and large doses of the colchicine group, Group showed a gradual reduction trend. Compared with the model group, the expression levels of Col-Ⅰ and Ⅲ mRNA in large, medium and small dose groups and colchicine group were significantly decreased (all P <0.05); compared with the colchicine group, Group Col-Ⅰ, III mRNA expression decreased (P <0.05). Compared with the model group, the expression of a-SMA in the Taibai alburnum wood large, medium and small dose group and the colchicine group decreased to a certain extent (all P <0.05). Compared with the colchicine group, The expression of a-SMA decreased (all P <0.05). [Conclusion] Taibai Alnus can reduce hepatic fibrosis by inhibiting the proliferation of collagen fibers and hepatic stellate cells in liver tissue.
