目的:建立HPLC同时测定杭菊中6种成分的方法。方法:采用Thermo AcclaimTM120 C18柱(250 mm×4.6 mm,5μm);甲醇-0.1%磷酸为流动相进行梯度洗脱;流速0.8 m L/min;柱温为30℃;检测波长为350 nm。结果:绿原酸、木犀草苷、3,5-O-双咖啡酰基奎宁酸、槲皮苷、木犀草素和芹菜素分别在0.0374~0.4488μg(r1=0.9997)、0.0400~0.4800μg(r2=0.9999)、0.0772~0.9264μg(r3=0.9999)、0.0143~0.1716μg(r4=0.9999)、0.0116~0.1392μg(r5=1.000)和0.0013~0.0150μg(r6=0.9997)范围内呈良好线性关系;加样回收率分别为99.2%、98.8%、99.7%、98.7%、98.1%和98.2%。结论:该方法简便、可靠、准确,可为杭菊的质量控制提供科学依据。
Objective: To establish a HPLC method for the simultaneous determination of 6 components in Hang Juzhong. METHODS: The gradient was eluted with a gradient of methanol-0.1% phosphoric acid as mobile phase on a Thermo AcclaimTM120 C18 column (250 mm × 4.6 mm, 5 μm) with a flow rate of 0.8 m L / min. The detection wavelength was 350 nm. RESULTS: The concentrations of chlorogenic acid, luteolin, 3,5-O-dicavanilylquinic acid, quercitrin, luteolin and apigenin were respectively 0.0374-0.4488μg (r1 = 0.9997), 0.0400-0.4800μg r2 = 0.9999), 0.0772 ~ 0.9264μg (r3 = 0.9999), 0.0143 ~ 0.1716μg (r4 = 0.9999), 0.0116 ~ 0.1392μg (r5 = 1.000) and 0.0013 ~ 0.0150μg The average recoveries were 99.2%, 98.8%, 99.7%, 98.7%, 98.1% and 98.2% respectively. Conclusion: The method is simple, reliable and accurate, which can provide scientific basis for the quality control of Hangzhou Chrysanthemum.
