目的:建立石龙清血颗粒含量测定超声提取的新方法,代替原标准中二步甲醇加热回流提取法,以缩短提取时间,节省试剂。方法:利用甲醇超声提取法、HPLC分析测定石龙清血颗粒中天麻素含量。以甲醇超声提取供试品,以十八烷基键合硅胶为填充剂,甲醇-磷酸盐溶液(0.1mol/L磷酸二氢钾和0.1mol/L磷酸二氢钠溶液等量混合)-水(1.5∶3∶95.5)为流动相,检测波长为210nm。结果:采用超声提取法,溶剂量大大减少,提取时间明显缩短,含量测定结果无明显差异。结论:该方法时间短,重现性好,精密度高,操作性强。
OBJECTIVE: To establish a new method for ultrasonic determination of the content of Shilongqingxue granule by replacing the two-stage methanol heating reflux extraction method in the original standard to shorten the extraction time and save the reagent. Methods: The content of gastrodin in Shilong Qingxue granule was determined by methanol ultrasonic extraction and HPLC analysis. The sample was extracted with methanol by ultrasound, and octadecyl-bonded silica gel was used as filler. The mixture of methanol-phosphate solution (0.1 mol / L potassium dihydrogen phosphate and 0.1 mol / L sodium dihydrogen phosphate solution) (1.5: 3: 95.5) as the mobile phase, detection wavelength of 210nm. Results: The ultrasonic extraction method greatly reduced the amount of solvent, extraction time was significantly shortened, no significant difference in the determination results. Conclusion: The method has short time, good reproducibility, high precision and good operability.
