为达到以结肠癌患者外周血树突状细胞(DC)在体外诱导抗结肠癌免疫反应的目的,以结肠癌细胞系LOVO肿瘤细胞的肿瘤抗原粗提物激活并经GM-CSF及IL-4联合刺激的结肠癌患者外周血树突状细胞(DC)体外能够诱导自体混合T淋巴细胞增殖、分化为CTL,该CTL及其上清液对LOVO肿瘤细胞均有强大的杀伤力,而对HepG2肿瘤,该CTL细胞及HOS-8603肿瘤细胞仅有微弱的细胞毒作用。结果表明结肠癌患者外周血DC体外能够诱导高效而特异抗结肠癌免疫反应。提示DC作为一新概念上的肿瘤疫苗可能在肿瘤治疗及预防中发挥重要作用
In order to achieve the purpose of inducing anti-colon cancer immune response in peripheral blood dendritic cells (DCs) from patients with colon cancer, the tumor antigens of colon cancer cell line LOVO tumor cells were activated with the crude extract of GM-CSF and IL-4. Peripheral blood dendritic cells (DCs) from patients with colon cancer that were co-stimulated could induce proliferation and differentiation of autologous mixed T lymphocytes into CTLs in vitro. The CTLs and their supernatants have a strong lethal effect on LOVO tumor cells, and they are also responsible for HepG2. Tumors, the CTL cells and HOS-8603 tumor cells had only weak cytotoxic effects. The results showed that peripheral blood DCs from patients with colon cancer can induce efficient and specific anti-colon cancer immune responses in vitro. It is suggested that DC as a new conceptual tumor vaccine may play an important role in cancer treatment and prevention
