It has been reported that biodegradable microspheres made of d, l-polylactic acid polymer sustained release of norethindrone for 6 or 3 months after single injections into primates. Blood norethindrone levels were dose-related. Lower doses had no significant effect on ovarian function, whereas higher doses maintained norethindrone levels sufficiently to suppress ovulation until 6 months. This article reports the effect of long-acting norethindrone microcapsule systems on fertility using a dose that does not inhibit ovulation. The microsphere system used contains 20% norethindrone, ranging in diameter from 100 to 125 μm, and sustained release of norethindrone for 3 months after a single intramuscular injection. Based on the evidence that microcapsules containing 25 to 100 mg of norethindrone, the normal menstrual cycle of baboons, the production of endogenous ovarian steroids
