目的探讨东莞地区汉族人群KCNJ11基因E23K位点多态性与2型糖尿病(type 2diabetes mellitus,T2DM)的关系。方法 T2DM患者50例为观察组,体检健康者50例为对照组,检测2组KCNJ11基因E23K位点多态性,比较2组空腹血糖(fasting plasma glucose,FPG)、体质量指数、空腹胰岛素(fasting insulin,FIns)和胰岛素抵抗指数(homeostasis model assessment of insulin resistance,HOMA-IR)。结果观察组FPG、体质量指数、FIns和HOMA-IR明显高于对照组(P<0.05);观察组E23K位点EE、EK、KK基因型频率分别为32.0%、48.0%和20.0%,E、K等位基因频率分别为56.0%和44.0%,对照组EE、EK、KK基因型频率分别为26.0%、56.0%、18.0%,E、K等位基因频率分别为54.0%、46.0%,2组比较差异无统计学意义(P<0.05);非条件logistic回归分析显示,FPG(OR=1.942,95%CI:1.013~2.283,P=0.000)、体质量指数(OR=1.122,95%CI:1.086~1.514,P=0.034)、FIns(OR=1.442,95%CI:1.213~1.701,P=0.013)和HOMA-IR(OR=1.763,95%CI:1.201~2.011,P=0.009)是影响T2DM发生的独立危险因素;FPG(OR=1.033,95%CI:1.012~1.832,P=0.032)、KK基因型(OR=1.482,95%CI:1.121~1.905,P=0.010)是肥胖的独立影响因素。结论未发现东莞地区汉族人群KCNJ11基因E23K位点多态性与T2DM发生有关,但KCNJ11基因E23K位点KK基因型可能是肥胖的危险因素。
Objective To investigate the association of KCNJ11 gene E23K polymorphism with type 2 diabetes mellitus (T2DM) in Han population in Dongguan area. Methods Twenty patients with T2DM were selected as the observation group and 50 healthy people as the control group. The E23K polymorphism of KCNJ11 gene was detected in two groups. Fasting plasma glucose (FPG), body mass index, fasting insulin fasting insulin, FIns) and homeostasis model assessment of insulin resistance (HOMA-IR). Results The FPG, body mass index, FIns and HOMA-IR in observation group were significantly higher than those in control group (P <0.05). The frequencies of EE, EK and KK at E23K in observation group were 32.0%, 48.0% and 20.0%, respectively The frequency of K and K alleles were 56.0% and 44.0% respectively. The frequencies of EE, EK and KK genotypes in control group were 26.0%, 56.0% and 18.0%, respectively. The frequencies of E and K alleles were 54.0% and 46.0% There was no significant difference between the two groups (P <0.05). The unconditional logistic regression analysis showed that body mass index (OR = 1.122, 95% (OR = 1.442, 95% CI: 1.213-1.701, P = 0.013) and HOMA-IR (OR = 1.763, 95% CI: 1.201-2.01 1, P = 0.034) (OR = 1.033, 95% CI: 1.012-1.832, P = 0.032) .KK genotype (OR = 1.482, 95% CI: 1.121-1.905, P = 0.010) was an independent risk factor for the occurrence of T2DM. Independent factors. Conclusion The E23K polymorphism of KCNJ11 gene in Han population in Dongguan area was not associated with T2DM. However, KK genotype at E23K locus in KCNJ11 gene may be a risk factor for obesity.
