目的:了解胰岛素样生长因子(insulin-like growth factor-1,IGF-1)对体外培养豚鼠肋软骨细胞分裂增殖及功能代谢的影响。方法:体外单层培养豚鼠肋软骨细胞,对照组培养液为无血清 DMEM,实验组在培养液 DMEM 中加入 IGF-1 使其终浓度分别为 10ng/ml、50ng/ml、100 ng/ml, 作用 6 天后, 检测细胞 DNA 含量和培养液中糖醛酸的含量。 结果:IGF-1 在 10~100ng/ml 浓度范围能明显增加培养软骨细胞的 DNA 及糖醛酸的含量,且以 50ng/ml 作用效果最明显,与对照组相比,有统计学意义(P<0.01)。结论:IGF-1 对体外培养肋软骨细胞有刺激,并以剂量依赖性方式影响细胞的增殖及功能代谢。
Objective: To investigate the effect of insulin-like growth factor-1 (IGF-1) on the division, proliferation and functional metabolism of cultured costal cartilage cells in guinea pigs. METHODS: Guinea pig costal cartilage cells were cultured monolayer in vitro. The control group was serum-free DMEM. The experimental group was given IGF-1 in culture medium DMEM to make the final concentrations of 10ng / ml, 50ng / ml, 100ng / ml, After 6 days, the content of DNA and the content of uronic acid in culture medium were detected. Results: The concentration of DNA and uronic acid in cultured chondrocytes was significantly increased in the concentration range of 10 ~ 100ng / ml, and the effect of 50ng / ml of IGF-1 was the most obvious, compared with the control group (P <0.01). CONCLUSION: IGF-1 stimulates cultured human chondrocytes in vitro and influences cell proliferation and functional metabolism in a dose-dependent manner.
